PaWOX3 and PaWOX3B Regulate Flower Number and the Lip Symmetry of Phalaenopsis.

Abstract

This article focuses on the roles of two WUSCHEL-related homeobox (WOX) genes, PaWOX3 and PaWOX3B, in the flower development of Phalaenopsis orchids. These genes are highly expressed during early floral bud stages and contribute to flower initiation and the regulation of flower number. Functional analyses, including virus-induced gene silencing (VIGS) and transgenic expression in Arabidopsis, indicate that PaWOX3/PaWOX3B regulate lateral organ differentiation and growth, and cooperate with the gene OAGL6-2—part of the L complex in the orchid P-code model—to maintain the symmetry of the orchid's distinctive lip structure. While single silencing of PaWOX3/PaWOX3B or OAGL6-2 does not disrupt lip symmetry, their combined knockdown leads to altered lip symmetry, highlighting their overlapping and complementary functions in orchid floral morphology.
