m5C and m6A modifications regulate the mobility of pumpkin CHOLINE KINASE 1 mRNA under chilling stress.

Abstract

This article investigates the species-specific mechanisms regulating the long-distance mobility of CHOLINE KINASE 1 (CK1) mRNA in plants, focusing on pumpkin (Cucurbita moschata) under chilling stress. Unlike Arabidopsis thaliana CK1 (AtCK1) mRNA mobility, which is mediated by transfer RNA–like sequences (TLS), pumpkin CK1 (CmoCK1) mRNA mobility is governed by both 5-methylcytosine (m5C) and N6-methyladenosine (m6A) RNA modifications. The study demonstrates that CmoCK1 mRNA moves from pumpkin rootstock to cucumber scion under chilling conditions, enhancing chilling tolerance by increasing phosphatidylcholine biosynthesis and activating cold-responsive genes. These findings reveal that mobile mRNAs and their regulatory mechanisms are not conserved across species and highlight m6A modification as a novel factor contributing to mRNA mobility in plants.
