Abstract

The article focuses on the role of TMEM41B, an endoplasmic reticulum (ER)-resident lipid scramblase, in the maturation and transport of glycosylphosphatidylinositol-anchored proteins (GPI-APs). It highlights that TMEM41B is essential for efficient transport of GPI-APs and transmembrane proteins from the ER to the Golgi and that its loss leads to accumulation of PGAP1 proteins in the ER, enhancing PGAP1-dependent deacylation of GPI-inositol. The study connects TMEM41B's regulation of ER membrane lipid integrity, particularly cholesterol and phosphatidylserine distribution, with the proper targeting and maturation of GPI-APs. These findings provide insights into the interplay between lipid scramblase activity, protein trafficking, and GPI anchor biosynthesis in eukaryotic cells.
