Nutritional importance of tryptophan for improving treatment in depression and diabetes.

Abstract
This article focuses on the essential amino acid tryptophan and its role in modulating central and peripheral serotonin synthesis, with implications for the treatment of depression and diabetes. Tryptophan is the sole precursor of serotonin, a neurotransmitter involved in psychiatric conditions such as depression and physiological processes including insulin secretion from pancreatic β cells. Stress-induced increases in glucocorticoids accelerate tryptophan degradation via the hepatic kynurenine pathway, reducing its availability for serotonin synthesis and potentially exacerbating both depression and diabetes. While preclinical and clinical studies show mixed results, evidence suggests that tryptophan supplementation may improve therapeutic outcomes, particularly when circulating tryptophan levels are depleted due to factors like stress or dietary insufficiency. Establishing guidelines for optimal circulating tryptophan levels could enhance treatment efficacy for these often comorbid diseases.
