Abstract

The article focuses on the development and validation of a green, selective, and accurate high-performance liquid chromatography (HPLC) method for the simultaneous separation and quantitation of omeprazole (OMEP), ondansetron (OND), and deflazacort (DEF) in spiked human plasma. Using an isocratic elution on an X Bridge C18 column with a methanol-ammonium acetate buffer mobile phase, the method achieved complete separation within six minutes and employed dexamethasone as an internal standard. Validation followed FDA bio-analytical guidelines, demonstrating acceptable linearity, accuracy, precision, selectivity, and stability across relevant concentration ranges for therapeutic drug monitoring. Additionally, four greenness assessment tools—including the National Environmental Method Index, Eco-scale, Analytical Greenness Metric Approach (AGREE), and Green Analytical Procedure Index (GAPI)—confirmed the method's environmentally friendly profile, highlighting its suitability for clinical and pharmacokinetic applications without the use of hazardous solvents.
