Knockout of OsSWEET15 Impairs Rice Embryo Formation and Seed-Setting.

Abstract

This article focuses on the role of the rice sugar transporter gene OsSWEET15 in seed development, demonstrating that its knockout significantly reduces rice fertility by causing about half of the spikelets to produce blighted or empty grains. OsSWEET15 is expressed in the embryo-surrounding endosperm tissues, specifically the embryo-adjacent scutellar aleurone layer (SAL) and embryo-adjacent region (EAS), where it likely facilitates sucrose export to support embryo development during the caryopsis formation stage. The study shows that in ossweet15 mutants, aborted grains fail to complete endosperm cellularization and embryo development, correlating with decreased sugar content, while empty grains are attributed to reduced pollen viability. Similar phenotypes in ossweet11 mutants and the complete sterility of double ossweet11/ossweet15 mutants indicate that both genes have overlapping functions in pollen development, caryopsis formation, and seed-filling in rice.
