Polyamidoamine dendrimer decorated graphene oxide as a pH-sensitive nanocarrier for the delivery of hydrophobic anticancer drug quercetin: a remedy for breast cancer.

Abstract

This article focuses on the development and evaluation of a polyamidoamine (PAMAM) dendrimer (generation 0) decorated graphene oxide (GO) nanocarrier for the targeted delivery of the hydrophobic anticancer drug quercetin (QSR) in breast cancer treatment. The GO-PAMAM nanohybrid was synthesized via covalent bonding and demonstrated higher QSR loading capacity and pH-sensitive, controlled drug release, with significantly greater release at acidic pH (4) compared to physiological pH (7.4). In vitro studies showed that QSR-loaded GO-PAMAM exhibited enhanced cytotoxicity against human breast cancer MDA MB 231 cells while maintaining biocompatibility with non-tumorigenic HEK 293T epithelial cells. These findings suggest that GO-PAMAM is a promising nanocarrier that improves QSR solubility, stability, and selective delivery, potentially enhancing therapeutic efficacy and reducing systemic toxicity in breast cancer therapy.
