Abstract

This article focuses on a clinical trial conducted in China investigating the effectiveness of clozapine as a sequential treatment for first-episode psychosis (FEP) after failure of one antipsychotic drug trial. The randomized, assessor-blind study involved 654 eligible patients aged 16 to 45, initially treated with one of five antipsychotics, with nonresponders rerandomized to olanzapine, amisulpride, or clozapine. Results showed that most patients responded to the first antipsychotic, with risperidone and amisulpride outperforming aripiprazole and perphenazine, while clozapine demonstrated superior efficacy in nonresponders compared to olanzapine and amisulpride.
