Fisetin alleviates chronic urticaria by inhibiting mast cell activation via MRGPRX2.

Abstract

This article investigates the therapeutic potential of fisetin, a natural flavonoid, in alleviating chronic urticaria (CU) by inhibiting mast cell (MC) activation via Mas-related G-protein coupled receptor X2 (MRGPRX2). Using murine models stimulated with ovalbumin (OVA) and substance P (SP), as well as SP alone, the study demonstrates that fisetin reduces urticaria-like symptoms, inflammatory mediator release, and MC degranulation. Molecular docking and surface plasmon resonance assays confirmed fisetin's binding to MRGPRX2, while cellular assays showed that fisetin suppresses MC activation by inhibiting calcium mobilization and downregulating phosphorylation of signaling proteins Akt, P38, NF-κB, and PLCγ. These findings suggest fisetin as a promising candidate for CU treatment through modulation of MRGPRX2-mediated MC pathways.
