Interferons and systemic lupus erythematosus: Pathogenesis, clinical features, and treatments in interferon-driven disease.

Abstract

This article focuses on the role of type I interferons (IFNs) in the pathogenesis, clinical manifestations, and treatment of systemic lupus erythematosus (SLE). Type I IFNs, particularly IFN-α, are implicated in driving SLE symptoms across multiple organ systems and are associated with disease activity, severity, and related autoimmune conditions such as Sjögren's syndrome and antiphospholipid syndrome. Although direct clinical measurement of type I IFN levels and the IFN gene signature (GS) is not routinely available, these biomarkers correlate with disease features and may predict treatment response. Therapeutic strategies targeting type I IFN pathways, including monoclonal antibodies like anifrolumab, have shown promise in reducing disease activity, marking a step toward precision medicine in SLE management.
