Berberine inhibits tumour growth in vivo and in vitro through suppressing the lincROR-Wnt/β-catenin regulatory axis in colorectal cancer.

Abstract
The article investigates the anti-tumour effects of berberine, a non-prescription isoquinoline alkaloid derived from Coptidis Rhizoma, on colorectal cancer (CRC) through epigenetic mechanisms. The study demonstrates that berberine inhibits CRC cell growth in vitro and tumour development in vivo by inducing cell cycle arrest and apoptosis. Mechanistically, berberine downregulates the long intergenic non-coding RNA lincROR, leading to inactivation of the canonical Wnt/β-catenin signalling pathway, which is frequently aberrantly activated in CRC. Overexpression of lincROR partially reverses berberine's suppressive effects on tumour growth and Wnt/β-catenin signalling, indicating that the lincROR-Wnt/β-catenin regulatory axis mediates berberine's anti-cancer activity. These findings suggest that targeting lincROR may enhance berberine's therapeutic potential in CRC management.
