Efficacy of a membrane concentration method combined with real-time PCR for detection of Giardia and Cryptosporidium in drinking water.

Abstract

This article focuses on the development and evaluation of a novel method combining membrane concentration and real-time polymerase chain reaction (PCR) for detecting the protozoan parasites Giardia and Cryptosporidium in drinking water. The method uses cellulose membrane filtration followed by nucleic acid extraction and real-time PCR targeting the gdh gene of Giardia and the 18S rRNA gene of Cryptosporidium, achieving detection limits of 0.926 and 0.65 copies/μL, respectively. Recovery rates exceeded 60% for Giardia and 38% for Cryptosporidium, with relative standard deviations below 2.3%, indicating good sensitivity, specificity, and reproducibility. This approach offers a cost-effective, less subjective alternative to traditional microscopy-based EPA methods, suitable for continuous environmental monitoring of waterborne protozoan pathogens in relatively clean drinking water sources.
