Abstract

This article focuses on the development and optimization of a rapid multiplex loop-mediated isothermal amplification (mLAMP) assay for the simultaneous detection of two common intestinal parasites, Entamoeba histolytica and Giardia duodenalis. Targeting the 18S rRNA gene of E. histolytica and the elongation factor 1 alpha (Ef1 α) gene of G. duodenalis, the assay distinguishes the parasites based on distinct melting temperatures within 30 minutes, demonstrating high analytical sensitivity (down to one femtogram of DNA) and specificity without cross-reactivity to other intestinal pathogens. The mLAMP assay operates under isothermal conditions, making it suitable for resource-limited and field settings, and shows potential applications in clinical diagnostics, environmental monitoring, and food quality assessment. Further validation with clinical samples is needed to confirm its utility in routine practice.
