Abstract
The article focuses on evaluating the use of nested polymerase chain reaction (PCR) targeting the flagellin gene (fliC) of Salmonella enterica serotype Typhi in urine samples for diagnosing typhoid fever. Conducted as an internal pilot study with 25 clinically diagnosed patients and 25 healthy controls in Cairo, Egypt, the nested PCR showed a sensitivity of 44% and specificity of 88%, with a positive predictive value of 78.57%. Blood culture demonstrated a low positivity rate of 4%. The study concludes that while nested PCR of urine samples has moderate sensitivity, its higher specificity indicates potential as a supplementary diagnostic tool for typhoid fever, warranting further validation before routine clinical use.
