Effects of Dextromethorphan on Nicotine-Induced Reward, Behavioral Sensitization, Withdrawal Signs, and Drug Seeking-Related Behavior in Rats.

Abstract

This article investigates the potential of dextromethorphan (DM), a widely used antitussive, to reduce nicotine dependence in rats. Using behavioral tests such as conditioned place preference (CPP) and withdrawal symptom scoring, the study found that both co-administration and post-treatment with DM suppressed nicotine-induced rewarding effects, drug-seeking behavior, withdrawal signs, and behavioral sensitization. Neurochemical analysis revealed that DM inhibited nicotine-induced increases in dopamine turnover specifically in the medial prefrontal cortex (mPFC), but not in the nucleus accumbens (NAc) or dorsal striatum (DS). These findings suggest that DM may have therapeutic potential for treating nicotine dependence, possibly through antagonism of α3β4 nicotinic receptors and modulation of mesocortical dopaminergic pathways.
