Optimizing Salmonella Detection in Beijing's Surface Water: Unveiling Rare Serotypes with a Modified Moore Swab (MMS) Method.

Abstract
This article focuses on the optimization and validation of a modified Moore swab (MMS) device for detecting Salmonella in large-volume surface water samples in Beijing. The MMS device demonstrated high efficiency (over 80% recovery) and a low detection limit (as few as 1 cell per 10 L), significantly improving Salmonella detection rates (62.86%) compared to conventional small-volume sampling (8.57%). Analysis of 28 Salmonella isolates from lakes, rivers, and irrigation water revealed 17 serotypes, mostly rare and associated with water wildlife, with the majority susceptible to tested antimicrobials. The study provides a reliable method and new data to support public health risk assessment of Salmonella contamination in Beijing's surface water systems.
