P32 Melanin confers protection against ultraviolet radiation-mediated mitochondrial DNA damage in skin of colour.

Abstract

This article investigates the protective role of melanin against ultraviolet radiation (UVR)-induced mitochondrial DNA (mtDNA) damage in human skin. By sequencing mtDNA from photoexposed forearm and photoprotected buttock samples of Black African and White Northern European volunteers, the study found significantly fewer somatic mtDNA mutations in Black skin compared to White skin. White skin showed an enrichment of cytosine to thymine (C>T) transitions, a known UVR damage signature, while Black skin exhibited a distinct thymine to guanine (T414G) transition associated with photoageing. These findings suggest that melanin provides protection against UVR-induced mtDNA damage and that mtDNA damage patterns vary between skin types.
