Novel genetic structures associated with adverse response to chemotherapy in breast cancer.

Abstract

This article focuses on identifying genetic variants and novel haplotypic structures associated with adverse responses to chemotherapy in breast cancer (BC) patients. Six significant single nucleotide polymorphisms (SNPs) located on PPCDC, NLRC5, STAM2, and TNFSF13B genes were found through genome-wide association studies (GWAS) to correlate with chemotherapy-induced toxicities such as neutropenia, leukopenia, cytotoxicity, and alopecia. Two genetic structures, GAG-TTAT and CC-CAACTCCCGTT, composed of these haplotypes, were linked to specific adverse chemotherapy responses and showed altered gene expression and gene-gene correlations in BC tumor tissues compared to normal tissues. The study suggests these genetic markers could form the basis of predictive panels for chemotherapy toxicity in BC, though validation in diverse populations is needed.
