Spotlight on Coenzyme Q10 in scopolamine-induced Alzheimer's disease: oxidative stress/PI3K/AKT/GSK 3ß/CREB/BDNF/TrKB.

Abstract

The article focuses on investigating the neuroprotective effects of Coenzyme Q10 (CoQ10) against scopolamine (Scop)-induced Alzheimer’s disease (AD)-like cognitive impairment in Wistar rats, emphasizing the modulation of oxidative stress and the PI3K/Akt/GSK-3β/CREB/BDNF/TrkB signaling pathway. Chronic co-administration of CoQ10 with Scop improved memory and learning deficits, reduced hippocampal amyloid beta (Aβ42) accumulation, oxidative damage markers (MDA, 8-OHdG), and restored antioxidant levels (GSH, SOD, catalase). Additionally, CoQ10 reversed Scop-induced dysregulation of key molecular components involved in AD pathogenesis, including decreased expression of APP and BACE-1, and normalized the phosphorylation states of PI3K, Akt, GSK-3β, and CREB, as well as mRNA levels of brain-derived neurotrophic factor (BDNF) and its receptor TrkB. These findings suggest that CoQ10 exerts neuroprotective effects by inhibiting oxidative stress, amyloid deposition, and modulating critical intracellular signaling pathways implicated in AD.
