The antiviral efficacies of small-molecule inhibitors against respiratory syncytial virus based on the F protein.

Abstract

This article focuses on identifying potential small-molecule inhibitors targeting the fusion protein (F protein) of respiratory syncytial virus (RSV) to treat RSV infection. Using virtual screening of FDA-approved drugs and molecular dynamics simulations, three compounds—Z65 (atracurium besylate), Z85 (homatropine methylbromide), and Z74 (sorbitan trioleate)—were identified and tested in vitro and in vivo. Z65 and Z85 demonstrated significant inhibition of viral replication and RSV-induced inflammation with low cytotoxicity, effectively reducing airway hyperresponsiveness and lung inflammation in RSV-infected mice, while Z74 showed limited protective effects. The study suggests that Z65 and Z85 are promising candidates for RSV treatment and provide a basis for developing new antiviral drugs targeting the RSV-F protein.
