Maintenance of the Golgi ribbon structure by the KASH protein Jaw1.

Abstract

This article focuses on the role of Jaw1, a Klarsicht/ANC-1/Syne/homology (KASH) protein and component of the linker of nucleoskeleton and cytoskeleton (LINC) complex, in maintaining the Golgi apparatus ribbon structure in mammalian cells. Jaw1 localizes to the endoplasmic reticulum and outer nuclear membrane and interacts with microtubules, contributing to the organization of the Golgi-derived microtubule network essential for Golgi ribbon integrity. Knockdown of Jaw1 disrupts the Golgi ribbon, causing fragmentation while preserving Golgi ministacks, and leads to a dispersed acetylated tubulin network, indicating impaired microtubule anchoring. The findings suggest Jaw1's involvement in Golgi morphology maintenance through microtubule organization and potentially calcium signaling via inositol 1,4,5-trisphosphate receptors, with implications for Golgi positioning and polarized transport.
