Abstract
The article focuses on the validation and reconstruction of the yeast artificial chromosome YAC-Mm-4q21LacO used to form functional human artificial chromosomes (HACs). Although the original yeast strain harboring YAC-Mm-4q21LacO was compromised, possibly due to contamination or prior strain use, the identity of the YAC that produced functional HACs in human cells was confirmed. The YAC was remade from its component parts and assembled in a host yeast strain, VL6-48N, with the new strain successfully forming functional HACs in human cell lines HT1080 and U2OS, consistent with prior findings.
