Abstract
This article focuses on the development and validation of a liquid chromatography–tandem mass spectrometry (LC–MS/MS) method for the simultaneous quantitative analysis of 30 flavonoid components in Jinqiancao granules, a traditional Chinese medicine derived from Lysimachia christinae Hance used to treat urinary calculi and related conditions. The method demonstrated high accuracy, specificity, reproducibility, and sensitivity, employing multiple reaction monitoring (MRM) mode to effectively distinguish flavonoid isomers. Chemometric analyses, including hierarchical cluster analysis (HCA), principal component analysis (PCA), and partial least squares discriminant analysis (PLS-DA), revealed significant batch-to-batch variability and identified Isoschaftoside, Vicenin-2, Schaftoside, and Myricitrin as key quality markers for Jinqiancao granules. These findings provide a scientific basis for improved quality control standards to ensure the consistency, safety, and efficacy of Jinqiancao granules in clinical use.
