Abstract
The article offers information on how cancer cells can undergo whole-genome doubling due to disruptions in cell cycle control pathways. Topics include the role of mitogenic signaling via cyclin-dependent kinases 4 and 6 (CDK4/6) in preventing cells from slipping back into the G1 phase; the role of the ubiquitin ligase APC/CCDH1 in ensuring DNA replication occurs only once per cell cycle; and the finding that stress-driven whole-genome doubling events can occur independently of the p53 pathway.
